“The Dark Beyond the Stars” by Frank M. Robinson is about an intergenerational ship searching for extraterrestrial life, starting to suspect it’s a hopeless mission. Just hopelessness and emotional dick-punches the whole way through.
“The Sparrow” by Mary Doria Russell is also gut-wrenching, but involves harm to children. It’s about a Jesuit mission to a new planet. The sequel gives some redemption.

You want Robin Hobb. She writes the most spectacular characters and then tortures them. Assassin’s Apprentice is probably where you should start.
